'Regulatory mechanisms of Syk and PARP1 in inflammation'

Abstract:

Induction of tissue regenerative macrophage to promote wound healing and liver regeneration

Shie-Liang Hsieh (Genomics Research Center, Academia Sinica, Taipei, Taiwan)

Macrophages play important roles in host defense against microbial infections, cancer progression, metabolic diseases, autoimmunity, and tissue repair. However the unique markers to distinguish macrophage subsets with distinct functions are far from satisfactory. The classical paradigm of M1 and M2 macrophage is unable to reflect the heterogenous phenotypes in M2 macrophage, thus ‘alternatively activated macrophage’ is used to reflect the diverse subsets of macrophages found in parasite infections, tumor progression, and tissue repair.

Recently, subsets of macrophages involved in tissue repair and resolution of inflammation was defined with several novel markers: including ATF3, CCL20, PPARgmma, PD-L1, PD-L2, etc (Immunity. 2016 Mar 15; 44(3): 450–462). However, the microenvironment and key factors for the induction of ‘tissue repair macrophage’ and ‘resolution macrophage’ is still unclear.

Here we demonstrated that the recombinant DcR3.Fc and HBD.Fc are potent inducers of ‘tissue repair macrophage’ and ‘resolution macrophage’, and their applications for tissue degeneration and repair will be addressed.
